Different isoforms of secretory PLA2 have been de- tected in nervous tissue, which participate in phys- iological and pathological processes. However, their specific roles and their subcellular localisation are still largely unknown. Using specific antibodies against sPLA2-IIA and sPLA2-V, we performed confocal im- munofluorescence experiments with the aim to analyse the subcellular distribution of these enzymes in PC-12 neuronal, U-251 astrocytoma and BV-2 microglia cell lines. In all these cell lines, both isoforms are present in the cytosol, however sPLA2-IIA is also found in mi- tochondria, and a large fraction of sPLA2-V is present in nuclei. When PC-12 cells are induced to differ- entiate into neuron-like cells, sPLA2-IIA appears to be concentrated in growth cones and tips of neurites, while the localisation of sPLA2-V remains unchanged. Treatment of astrocytoma or microglial cells with LPS enhances the expression of sPLA2-V and, to a smaller extent, of sPLA2-IIA. These results indicate that the two enzyme isoforms have a different subcellular lo- calisation in neural cells likely reflecting specific cel- lular functions, despite their high structural homology.
