The aim of this research was to compare Stearoyl-CoA desaturase (SCD) gene expression, an enzyme involved in lipid biosynthesis, in normal and double muscled Italian Marchigiana beef cattle. The Marchigiana is a cattle breed with significant somatic development, characterized by a high growth capacity and outstanding precocity. The Marchigiana breed, in Italy, is widespread throughout the Marche, Lazio, Abruzzo and Campania areas. Approximately 50000 heads are enrolled with the Herd-Book. The myostatin gene is a negative control factor in muscle growth. When mutation in myostatin gene is present, produces a complete lack of protein synthesis causing an abnormal growth in skeletal muscle in carrier animals. The mutation is a G to T transversion at nucleotide 874 in exon 3, known as E291X. Three animals without mutation (normal) and six with mutation (3 homozygous, 3 heterozygous) were analysed. Total RNA was extracted from three different skeletal muscle (Semitendinosus, Psoas major and Longissimus thoracis lumborum) representative of three different metabolisms (oxidative, glycolitic and intermediate). Expression level of SCD gene was performed in Real Time PCR using as reference the 18S rRNA gene. In animal carrying mutation the results, related to all tested muscle, showed low expression level of SCD gene as compared with normal animals (1.00 vs 1.76).
