The incubation of skeletal muscle in the presence of theophylline and Imidazole causes an increase in cyclic AMP levels while it has no effect on cyclic GMP concentration. Tetanic stimulation does not change the levels of the cyclic nucleotides being measured at the end of stimulation in muscles treated with normal Ringer solution or in the presence od Theophylline. In the presence of Imidazole, there is a further increase in cyclic AMP levels. An inverse correlation between cyclic AMP concentration and the amount of isotonic contraction has been demonstrated.
