Investigations on Verticillium wilt of olive tree were conducted for three years (1997-99) in two Umbrian orchards. Field observations confirmed that climatic and agrnomic factors, weak virulence of fungal strains, defence reactions of the plant determine fluctuations in the presence of the pathogen during the year and influence the natural recovery of some infected trees. Surveys on fungal microflora occurring in olive phylloplane showed that Coniothyrium sp. was isolated most frequently among other fungal species. DAS-ELISA was tested for its suitability in the detection of V.dahliae on olive tree. Pure cultures of some V.dahliae isolates reacted positively whereas naturally infected twigs gave negative responsed. These negative reactions are probably due to the low concentration of the pathogen in infected tissues or to the difficult extraction of antigens from infected samples.
